RESUMO

Uncompetitive inhibition is an effective strategy for suppressing dysregulated enzymes and their substrates, but discovery of suitable ligands depends on often-unavailable structural knowledge and serendipity. Hence, despite surging interest in mass spectrometry-based target identification, proteomic studies of substrate-dependent target engagement remain sparse. Herein, we describe the Thermal Shift Assay with ATP and RNA (TSAR) as a template for proteome-wide discovery of substrate-dependent ligand binding. Using proteomic thermal shift assays, we show that simple biochemical additives can facilitate detection of target engagement in native cell lysates. We apply our approach to rocaglates, a family of molecules that specifically clamp RNA to eukaryotic translation initiation factor 4A (eIF4A), DEAD-box helicase 3X (DDX3X), and potentially other members of the DEAD-box (DDX) family of RNA helicases. To identify unexpected interactions, we optimized a target class-specific thermal denaturation window and evaluated ATP analog and RNA probe dependencies for key rocaglate-DDX interactions. We report novel DDX targets of the rocaglate clamping spectrum, confirm that DDX3X is a common target of several widely studied analogs, and provide structural insights into divergent DDX3X affinities between synthetic rocaglates. We independently validate novel targets of high-profile rocaglates, including the clinical candidate Zotatifin (eFT226), using limited proteolysis-mass spectrometry and fluorescence polarization experiments. Taken together, our study provides a model for screening uncompetitive inhibitors using a systematic chemical-proteomics approach to uncover actionable DDX targets, clearing a path towards characterization of novel molecular clamps and associated RNA helicase targets.

AIM: Mesothelioma is associated with asbestos exposure. In this case series, we present 166 cases of individuals who had substantial asbestos exposure to cosmetic talc products as well as some who had potential or documented additional exposures to other asbestos-containing products and who subsequently developed mesothelioma. METHODS: Data were gathered for all subjects referred to an occupational and environmental medicine specialist as part of medicolegal review. Years of total cosmetic talcum powder usage was noted as well as the latency from the onset of talcum powder use to the mesothelioma diagnosis. Alternate asbestos exposure in addition to the exposure from cosmetic talc was categorized as none, possible, likely, and definite. RESULTS: In 122 cases, the only known exposure to asbestos was from cosmetic talc. For 44 cases, potential or documented alternate exposures in addition to the cosmetic talc were described. CONCLUSION: Cumulative exposure to asbestos leads to mesothelioma; for individuals with mixed exposures to asbestos, all exposures should be considered. Use of cosmetic talc is often overlooked as a source of asbestos exposure. All individuals with mesothelioma should have a comprehensive history of asbestos exposure, including cosmetic talc exposure.

The cichlid fishes of Lake Malawi are the paramount example of adaptive radiation in vertebrates. Evidence of their astounding diversity is perhaps most visible in their adaptations for obtaining food; the genus Labeotropheus, due to their prominent snouts, are an interesting example of an extreme adaptation for feeding. Two different body types are found in this genus: a deep-bodied form (e.g., L. fuelleborni) found most often in turbulent shallow water; and a slender bodied form (e.g., L. trewavasae) found in structurally-complex deep water habitats. Here we test the hypothesis that L. trewavasae should suffer a loss in fitness, measured as growth rate, if raised in turbulence; additionally, we examined growth and morphology of L. fuelleborni and L. fuelleborni x L. trewavasae hybrids under these conditions. We did find the predicted loss of fitness in turbulent-raised L. trewavasae, but found no loss of fitness for L. fuelleborni in either condition; hybrids, due to an unusual morphology, performed better in turbulent as opposed to control conditions. Fitness in turbulent conditions was dependent upon morphology, with deeper bodies and upturned neurocrania allowing a greater growth rate under these conditions. Directional selection on morphology was crucial in the evolution of morphology in the Labeotropheus.
